Operation method
Morphological observation experiments on the growth manifestations of bacteria Move Staphylococcus albicans agar plate cultures, Escherichia coli and dysentery bacillus semi-solid cultures, Escherichia coli slant cultures, Staphylococcus aureus, Bacillus subtilis, and streptococcus broth cultures. 1, Growth performance in liquid media:
(1) turbid growth: the liquid is uniformly turbid, or there is a small amount of precipitation. Most bacteria are so.
(2) precipitation growth: the liquid is clear, bacteria are mainly concentrated in the bottom of the tube to form a precipitate. This is true for streptococci.
(3) Surface growth: the liquid is clear and the bacteria grow mainly on the surface of the liquid to form a bacterial film. This is true for specialized aerobic bacteria.
2, the growth performance in solid medium
(1) Bacterial moss: bacteria form a layer of moss on the surface of the medium.
(2) colony: a single bacteria in the solid culture medium breeding group called colony. Observe the color, size, morphology, gloss, edge, hemolysis, etc. can be identified colonies; can be purified by separating the colonies on the agar plate; in the hygiene test, the number of colonies can be counted to understand the content of bacteria in the test product.
3、Growth performance in semi-solid culture medium
(1) Linear growth: Bacteria without flagellum can not move, just grow along the puncture line, and the boundary around the puncture line is clear.
(2) Foggy (brush-like) growth: Bacteria with flagella grow diffusely along the puncture line due to their ability to move, and the boundary around the puncture line is blurred.
