Certain bacteria secrete the enzyme peroxynitrite, which causes hydrogen peroxide to produce water and primordial oxygen, followed by the formation of oxygen molecules that appear as bubbles.
Operation method
Bacterial catalase assay
Principle
Certain bacteria secrete the enzyme peroxynitrite, which causes hydrogen peroxide to produce water and primordial oxygen, followed by the formation of oxygen molecules that appear as bubbles.
Materials and Instruments
H2O2 solution Move I. Experimental reagents: Caveat 1. Hydrogen peroxide is 3%, application of 30% concentrated H2O2 should be diluted 10 times. 2. Tests should not be done on culture media. 3. positive and negative strains should be used as controls for each test.Positive strains: Neisseria spp. are positive, Streptococcus spp. Kingella spp. are negative. 3% H2O2 solution, kept in a brown bottle at 4°C in a dark place.
II.Experimental method:
Pick the colonies on the solid medium first, place them on a clean slide, and then add 1-2 drops of 3% hydrogen peroxide solution. If it produces a lot of bubbles within 1min, it is positive, and if it does not produce bubbles, it is negative.
