Principle

Experimental principle of tungstic acid method: sodium tungstate mixed with sulfuric acid to generate tungstic acid, tungstic acid can be combined with blood proteins to generate protein salt precipitation, and centrifugation to remove the protein. Experimental principle of trichloroacetic acid method: blood proteins can be combined with trioxoacetic acid to generate insoluble protein salt precipitate, the protein is removed by centrifugation, and the supernatant is protein-free hemofiltrate.

I. Tungstic acid method

Experimental reagents:

1. 100g/L sodium tungstate solution: weigh 100g of sodium tungstate, dissolve in water and add water to 1000ml

2. 1/3M sulfuric acid: take 2 parts of equivalent sulfuric acid calibrated by yourself and mix with 1 part of water.

Experimental operation:


Slowly put whole blood into distilled water with a pipette, mix thoroughly after each addition of reagents, centrifuge after 10 minutes of rest, and take the supernatant for spare.

Trichloroacetic acid method

Experimental reagent: 100 g / L trichloroacetic acid solution

Experimental operations: take a serum or plasma, 9 TCA, add serum or plasma (add while shaking). Sufficiently mixed after centrifugation, take the above method for backup.

Caveat

I. Tungstic acid method1. When there is excess oxalate in the anticoagulated blood, the protein precipitation is incomplete, and the precipitate cannot become dark brown, at this time, 100g/L sulfuric acid can be added drop by drop, and then filtered by shaking well until the precipitate becomes dark brown.

2. When the protein filtrate is not transparent, the protein precipitate and filtrate should be poured back into the test tube, and sulfuric acid should be added according to the method shown in 1, so that the protein is completely precipitated.

3. This filtrate is suitable for glucose, creatinine, creatine, uric acid and other measurements.Second, trichloroacetic acid method1, the filtrate obtained from this method, more acidic, suitable for the determination of strong acidic dissolved substances, such as calcium, phosphorus, etc.. Phosphorus, etc.

2. It is not suitable for the quantitative determination of alkaline reduction of blood sugar.

3. Trichloroacetic acid is easy to deliquesce, pay attention to preservation.
