UltraBio™ GST-tag Purification Resin provides a simple, rapid, and highly efficient method for purifying GST-tagged proteins from a broad host such as bacteria, mammals, insects, Baculovirus, and yeast.Glutathione-S-transferase is often used as a tag for protein expression and purification applications. It is typically fused to the N terminus of a recombinant protein. GST tag has a high affinity for glutathione which is immobilized on UltraBio™ GST-tag Purification Resin. Therefore, the UltraBio™ GST-tag Purification Resin can be used to purify GST-tagged recombinant proteins from a crude lysate, while most other proteins in the lysate will not bind to the resin, or bind only weakly. The GST-tagged protein purified under mild conditions without denaturation usually maintains bioactivity and can be used for protein structure and function analysis, antibody preparation, protein interaction studies, protein-nucleic acid interaction studies, etc.GST can act as a chaperone to facilitate protein folding and increases the solubility of GST-tagged recombinant proteins, which helps maintain the full bioactivity of the target protein. Many eukaryotic proteins that are insoluble when expressed inu2002E. coli are at least partially soluble when expressed as a GST fusion protein.GST tag can be removed from the recombinant protein if a protease cleavage site of PreScission Protease, TEV Protease, or Thrombin, is engineered between the GST tag and the target protein.UltraBio™ GST-tag Purification Resin is based on highly crossed-linked 6% agarose gel (Sepharose CL-6B) on which GSH is immobilized by the optimized arm and GSH coupling technology. Compared to other products with similar functions, UltraBio™ GST-tag Purification Resin exhibits lower non-specificity, stronger resistance to pressure, and stabler coupling of GSH to the resin.GST-tagged proteins should be purified under non-denaturing conditions. Proteins in inclusion bodies dissolved with 8M urea or 6M guanidine hydrochloride should be dialyzed to remove the denaturants and then renaturated before protein purification with this product.UltraBio™ GST-tag Purification Resin has a particle diameter of 45-165μm. It can withstand pressure up to 0.025MPa (~5.8psi). The recommended flow rate is 0.5ml/min when using a fixed flow rate for protein purification.UltraBio™ GST-tag Purification Resin binds GST-tagged protein with high capacity and high specificity. Per milliliter of gel can bind 5-6mg of GST, 8-12mg of GST-tagged protein with a molecular weight of 50kD, or 12-18mg of 100kD GST-tagged protein. The maximum binding capacity in actual use depends on the molecular weight and characteristics of the protein to be purified. The larger the molecular weight, the greater the maximum binding capacity. For proteins with the same molecular weight, the maximum binding capacity of this resin might be different due to different characteristics of the proteins.This product is stored in 20% ethanol. A total volume of 10ml consists of 5ml gel and 5ml liquid. It is advisable to fully resuspend the gel prior to use.Using 1ml of this product, up to 2.5-3.0mg of GST protein and up to 4-6mg of GST-tagged recombinant proteins with a molecular weight of 50kD can be purified.
